Description
DNA-midi™ GT Plasmid DNA Extraction Kit (Gravity) from iNtRON Biotechnology is designed for efficient midi-scale isolation and purification of high-quality plasmid DNA from bacterial cultures.
The kit combines an optimised alkaline lysis buffer system with gravity-flow columns packed with anion-exchange resin. Following bacterial cell harvesting, resuspension, alkaline lysis and neutralisation, the clarified lysate is applied to the column. Plasmid DNA binds to the anion-exchange resin under suitable salt and pH conditions, while contaminants are removed through washing. The purified plasmid DNA is subsequently eluted, precipitated and recovered for downstream use.
DNA-midi™ GT can process up to 60 mL of bacterial culture containing a high-copy plasmid or up to 120 mL of culture containing a low-copy plasmid. The system can provide up to approximately 100 µg of purified plasmid DNA, although actual recovery depends on the bacterial strain, culture conditions, plasmid copy number and culture volume.
The gravity-flow design does not require centrifugation to move the sample through the purification column. The complete midi-preparation workflow can be completed in approximately 120 minutes and provides purified plasmid DNA suitable for a broad range of molecular biology applications.
