Description

DNA-spin™ Plasmid DNA Purification Kit from iNtRON Biotechnology provides a rapid and efficient method for isolating high-quality plasmid DNA from bacterial cultures using alkaline lysis and silica-membrane spin-column technology.

The kit is suitable for purification of both high-copy and low-copy plasmid DNA. Following bacterial cell resuspension, alkaline lysis and neutralisation, plasmid DNA selectively binds to the silica membrane under high-salt conditions. Contaminants are removed through washing steps, and purified plasmid DNA is then eluted in a small volume of low-salt elution buffer.

The included Lysis Viewer allows the pH changes occurring during cell lysis and neutralisation to be observed visually. This helps users confirm adequate mixing and reduces common handling errors associated with incomplete cell lysis or neutralisation.

The kit can provide up to 20 µg of highly purified plasmid DNA, while the specialised silica membrane has a maximum DNA-binding capacity of approximately 35 µg. It supports purification of both high-copy and low-copy plasmids, including plasmid DNA up to approximately 30 kb.

Purified plasmid DNA is free from major protein, RNA and chromosomal DNA contaminants and is ready for use in downstream molecular biology procedures without additional precipitation, concentration or desalting.
