Description

Fast DNA-spin™ Plasmid DNA Purification Kit from iNtRON Biotechnology is designed for rapid separation and purification of high-quality plasmid DNA from overnight E. coli cultures using alkaline lysis and silica-membrane spin-column technology.

The kit provides a simplified plasmid purification workflow that can be completed in approximately 10 minutes, excluding bacterial culture preparation. It is suitable for processing 1–4 mL of overnight E. coli culture and eliminates the need for an ice-incubation step.

Unlike conventional plasmid purification procedures, centrifugation of the neutralised bacterial lysate before column loading is not required. Following cell resuspension, alkaline lysis and neutralisation, the prepared lysate is applied to the silica-membrane spin column. Plasmid DNA binds to the membrane, contaminants are removed by washing, and purified DNA is subsequently eluted for immediate downstream use.

The included Lysis Viewer enables visual monitoring of the pH changes occurring during bacterial cell lysis and neutralisation. This provides a convenient indication of correct buffer mixing and helps reduce errors caused by incomplete lysis or neutralisation.

Purified plasmid DNA is suitable for restriction enzyme digestion, DNA sequencing, library screening, ligation and transformation, in vitro translation and transfection of robust cell lines.
