Description

i-Pfu™ DNA Polymerase with Buffer, dNTPs is a high-fidelity, thermostable DNA polymerase from iNtRON Biotechnology designed for PCR applications requiring accurate DNA synthesis. The enzyme is derived from Pyrococcus furiosus DNA polymerase and catalyses DNA synthesis in the 5′→3′ direction.

i-Pfu™ DNA Polymerase possesses 3′→5′ exonuclease proofreading activity, allowing incorrectly incorporated nucleotides to be removed during DNA synthesis. This produces PCR products with greater sequence accuracy than conventional Taq DNA polymerase and makes the enzyme particularly suitable for cloning, expression studies, mutagenesis and amplification of templates intended for DNA sequencing.

The polymerase produces blunt-ended amplification products and is recommended for high-fidelity amplification of genomic DNA and cDNA targets up to 10 kb. It can be used with a variety of DNA templates, including cloned fragments, plasmid DNA, phage DNA, cDNA and mammalian genomic DNA.

The product is supplied at a concentration of 2.5 U/µL together with 10X PCR Buffer containing Mg²⁺, 10X Mg²⁺-free PCR Buffer, dNTP mixture and a separate MgCl₂ solution. These components provide flexibility for optimization of magnesium concentration and other PCR conditions.
