01 Description, applications & specifications
Description
i-StarMAX™ II DNA Polymerase with Buffer, dNTPs is a Hot-Start, enhanced-fidelity DNA polymerase blend from iNtRON Biotechnology. It combines modified i-StarTaq™ Hot-Start DNA Polymerase with a proofreading DNA polymerase to provide high specificity, fidelity and amplification yield.
The Hot-Start polymerase component remains inactive at lower temperatures and becomes active during thermal cycling. This helps minimize premature primer extension, primer-dimer formation and nonspecific amplification during reaction preparation and the initial heating stages.
The proofreading polymerase component improves amplification accuracy, while the processivity of the Taq component supports efficient amplification of both short and long DNA fragments. The combined system is recommended for relatively rapid, high-fidelity amplification of genomic DNA and cDNA targets up to 15 kb.
i-StarMAX™ II DNA Polymerase is particularly useful for problematic template-and-primer systems, GC-rich or structured sequences, long-range PCR and multiplex PCR. Recommended extension time is approximately 30 seconds to 1 minute per kilobase, depending on target length and reaction conditions.
The enzyme is supplied at a concentration of 5 U/µL with 10X PCR Buffer containing Mg²⁺, 10X Mg²⁺-free PCR Buffer, dNTP mixture and a separate Mg²⁺ solution. These components allow flexible optimization of magnesium concentration for different templates and primer systems.
Key Features: Hot-Start PCR capability • Enhanced amplification fidelity • High specificity and sensitivity • Reduced nonspecific amplification • Reduced primer-dimer formation • High amplification yield • Long-range PCR up to 15 kb • Suitable for difficult and GC-rich templates • Compatible with multiplex PCR • Flexible magnesium optimization • Supplied with PCR buffers, dNTPs and Mg²⁺
