Description

RedSafe™ Nucleic Acid Staining Solution from iNtRON Biotechnology is a 20,000X concentrated fluorescent stain for visualisation of DNA and RNA during agarose gel electrophoresis.

RedSafe™ is designed as a safer alternative to traditional ethidium bromide staining while providing comparable nucleic acid detection sensitivity. The stain emits green fluorescence when bound to DNA or RNA and can be incorporated directly into the agarose gel before casting.

When bound to nucleic acids, RedSafe™ has fluorescence excitation maxima centred at approximately 309 nm and 419 nm, together with an additional visible excitation at approximately 514 nm. Its fluorescence emission maximum is centred at approximately 537 nm.

The stain is suitable for detecting double-stranded DNA and single-stranded RNA in agarose gels. Manufacturer testing shows detection of PCR products, genomic DNA, DNA markers and total RNA under UV illumination.

The standard pre-staining procedure uses 5 µL of RedSafe™ 20,000X concentrate per 100 mL of prepared agarose solution. The stain is mixed into the molten agarose before the gel is cast, eliminating the need for a separate post-electrophoresis staining step.

RedSafe™ can be used for routine PCR product analysis, restriction-fragment analysis, plasmid DNA analysis, genomic DNA analysis, RNA analysis and isolation of DNA fragments for downstream cloning workflows.

Recommended Pre-Staining Procedure

  1. Prepare 100 mL of 0.8–3% agarose solution in the appropriate electrophoresis buffer.
  2. Heat until the agarose is completely dissolved and the solution is clear.
  3. Add 5 µL of RedSafe™ 20,000X to the 100 mL agarose solution.
  4. Swirl gently to mix while avoiding the formation of bubbles.
  5. Pour the agarose into the prepared casting tray and insert the comb.
  6. Allow the gel to solidify, load the samples and perform electrophoresis.
  7. Visualise the nucleic acid bands using a compatible UV or fluorescence imaging system.

Important: Repeated melting of agarose gels containing RedSafe™ may reduce staining sensitivity. For optimal detection, the manufacturer recommends keeping the gel thickness below 0.5 cm.

Safety: Although RedSafe™ is positioned as a safer alternative to ethidium bromide, it may cause skin or eye irritation. Wear suitable gloves and eye protection, consult the safety data sheet, and dispose of the stain and stained gels according to applicable institutional and local requirements.
